Once a creature is considered blocked, it will only deal damage to blocking creatures, even if that creature is killed before damage. If your opponent blocks a creature without trample and you kill the blocker before damage, you will not be able to assign damage to your opponent.

This has actually been true since Alpha; blocked creatures have never been able to deal damage to the defending player unless they have trample.

The same rules apply. As soon as the creature is blocked, it will no longer deal damage to the defending player, regardless of whether the creature blocking it is still alive to receive damage.

Defending creature does damage to attacking creature?

No. The defending creature will not be on the battlefield during the Combat Damage Step so it will neither deal nor recieve damage.
