If I have out an Outrider en-Kor and a 1/1 creature, and my opponent casts a spell that deals 5 dmg to target creature on my Outrider en-Kor; can I transfer all 5 of the damage to my 1/1 creature or can I only use the ability once?

Outrider en-Kor reads:
"2/2 Flanking.
(0): the next 1 dmg that would be delt to Outrider en-Kor this turn is dealt to target creature you control instead."

You can play the ability as often as you want, whenever you want. You can play the ability five times to move all five damage of the spell to the 1/1.

yes, you can, but only because the 5 damage is simultaneous.
if you had, for example, 5 grapeshot copies all targeting outrider, the 1/1 would absorb 1 of them, but by the time the other 4 resolve, the 1/1 is dead and can take no more damage, so the outrider would die.

you can also use the ability even if no damage is being dealt
to pump up your angelic protector/task force to give it
infinitely high toughness
